How can i prepare ampicillin and kanamycin stock solution?
You got to prepare stock solutions of Ampicillin (100 mg/mL) and Kanamycin (50 mg/mL). Then add 1 uL of Ampicilln or Kansmycin in 1 mL medium (it will be 1:1000 diluted). Working concentration of the Ampicillin and Kanamycin is 50-100 and 25-50 ug/mL respectively.
You got to prepare stock solutions of Ampicillin (100 mg/mL) and Kanamycin (50 mg/mL). Then add 1 uL of Ampicilln or Kansmycin in 1 mL medium (it will be 1:1000 diluted). Working concentration of the Ampicillin and Kanamycin is 50-100 and 25-50 ug/mL respectively.
If you’ll be preparing plates with a final concentration of 100 ug/mL ampicillin, you should make a stock solution of 100,000 ug/mL (100 mg/mL).
1. Mix 1 g Amp powder into 10 mL of d H2O. 2. Vortex until powder is thoroughly dissolved. 3. Sterilize using 0.22-μm sterile filter. 4. Aliquot 1 mL of Amp solution per 1.5 mL tube and store at -20 °C.
Dilute your antibiotic into your LB medium at 1:1,000. For example, to make 100 mL of LB/ampicillin growth media, add 100 μL of a 100 mg/mL ampicillin stock.
Ampicillin : add 1ml ampicillin (at 100mg/ml) per liter of agar to obtain a final concentration of 100ug/ml.
Kanamycin : add 1ml kanamycin stock (at 50mg/ml) per liter of agar to obtain a final concentration of 50ug/ml.
Tetracycline : add 1ml tetracycline stock (at 15mg/ml) per liter of agar to obtain a final concentration of 15ug/ml.
Chloramphenicol :add 1ml chloramphenicol stock (at 25mg/ml) per liter of agar to obtain a final concentration of 100ug/ml.
How to determine volumes to use to obtain a certain concentration:
Use the following formula to determine amount of stock solution to use for any amount.
C1V1 = C2V2 C = concentration; V = volume
Example: You have a stock solution of A at 100mg/ml. You want 150 ml at 100ug/ml. Use the C1V1 for what you want, and the C2V2 for what you have. Just plug in the numbers:
(100ug/ml)(150ml) = (100000ug/ml)(x) 15000 = 100000x x = 0.15ml, or 150ul
Remember that you have to convert mg/ml to ug/ml, which you can do by multiplying by 1000.
