The anti-tumor activity of vinorelbine is believed to be mainly inhibited by interfering with tubulin in metaphase mitosis. Similar to other vinca alkaloids, vinorelbine can also interfere with: 1) the metabolism of amino acids, cyclic AMP and glutathione; 2) calmodulin-dependent calcium ion transport ATPase activity; 3) cellular respiration; 4) nucleic acid and lipid biosynthesis. In mouse intact embryo culture, vinorelbine inhibited microtubule formation at the same concentration (2uM), including blocking the metaphase division of cells. These data indicate that vinorelbine has a relatively selective effect on microtubules in metaphase of mitosis.

It can reduce the increase of serum AST and ALT in the animal acute liver injury model caused by TAA (thioacetamide) and CC14 (carbon tetrachloride), and inhibit the accumulation of triglycerides caused by chronic liver injury model; it can promote liver glycogen synthesis, inhibit the increase of cholesterol, and help to restore the serum albumin/globulin ratio.

It antagonizes histamine and acetylcholine, relieves bronchospasm and has an anti-asthmatic effect; it has a strong inhibitory effect on the cough center; it causes increased secretions, thins thick sputum, reduces sputum viscosity, and makes it easier to cough up; it has a similar effect to aminophylline in relaxing bronchial smooth muscles; it increases the electrical stimulation threshold of the cerebral cortex, inhibits synaptic conduction of electrical stimulation and the spread of epileptic electricity.

Generally, when the dosage is lower than 10 mg per kg of body weight per day, the side effects are mild or not obvious. When the dosage exceeds 10 mg per kg of body weight per day, loss of appetite, dizziness, tinnitus, general fatigue, nausea, etc. may be seen. The causal relationship between the above reactions and the treatment itself has not yet been determined.
