Function and Efficacy

This product belongs to the azole antifungal drug, and it can inhibit the process of Candida albicans transforming from spores to invasive hyphae. This product has a broad-spectrum antifungal activity, and has good antibacterial effects on Epidermophyton, Trichophyton, Aspergillus, Chromatid fungi, Cryptococcus and Candida. It also has certain antibacterial activity against Sporothrix schenckii, Blastomyces dermatitidis, Coccidioides and Histoplasma. This product has poor effects on Aspergillus, some dark spores, Mucor and other genus. This product inhibits the biosynthesis of ergosterol, the main steroid of fungal cell membrane, by interfering with the activity of cytochrome P-450, damaging the fungal cell membrane and changing its permeability, resulting in leakage of important intracellular substances. This product can inhibit the biosynthesis of triglycerides and phospholipids of fungi, inhibit the activity of oxidases and peroxidases, and cause the accumulation of hydrogen peroxide in cells, leading to cell submicrostructure degeneration and cell necrosis.

Appearance

This product is white tablets.

Indication

It is used to prevent and treat oral and esophageal Candida infections in immunosuppressed patients. However, due to its poor oral absorption, poor efficacy in treating deep fungal infections, and common adverse reactions, it is now rarely used and is only used topically.

Usage and Dosage

Oral administration: 0.25-1g at a time, 0.75-3g per day. Children: 20-60mg/kg per day, divided into 3 doses.

Adverse Reactions

1. Common gastrointestinal reactions after oral administration, generally after starting to take the drug, loss of appetite, nausea, vomiting, abdominal pain, diarrhea, etc., usually need to stop taking the drug in severe cases. 2. Hepatotoxicity: Since most of this product is metabolized in the liver, liver damage may occur, causing elevated serum bilirubin, alkaline phosphatase and aminotransferase, which can be restored after stopping the drug. 3. Occasionally, temporary neuropsychiatric abnormalities may occur, manifested as depression, hallucinations and disorientation. Once such reactions occur, treatment must be discontinued.

Precautions

It is contraindicated for patients with liver dysfunction, granulocytopenia, adrenal cortex insufficiency and allergy to this product.

Drug Interactions

This product has no synergistic effect with nystatin, amphotericin B and flucytosine against Candida albicans.
