Function and Efficacy
This product is a formyl derivative of tetrahydrofolate, mainly used for the rescue of high-dose methotrexate and other folic acid antagonists. The main function of methotrexate is to bind to dihydrofolate reductase, blocking the conversion of dihydrofolate into tetrahydrofolate, thereby inhibiting DNA synthesis. After entering the body, this product is converted into tetrahydrofolate by tetrahydrofolate reductase, which can effectively counteract the toxic reaction caused by methotrexate, but has no obvious effect on the existing methotrexate neurotoxicity.

Appearance
This product is off-white or light yellow tablets.
Indication
1. Used as an antidote for folic acid antagonists (such as methotrexate, pyrimethoprim or trimethoprim). 2. Used to prevent severe toxic effects caused by overdose or high-dose treatment with methotrexate. 3. Also used to treat megaloblastic anemia caused by folic acid deficiency. 4. Used in combination with 5-fluorouracil for advanced colon and rectal cancer.
Usage and Dosage
1 Oral administration of this product as a "rescue" therapy for methotrexate, the dosage is best determined based on the blood drug concentration. The commonly used dose is 5-15 mg, once every 6-8 hours, for 2 consecutive days. 2 As an antidote for pyrimethamine or trimethoprim, the daily dose is 5-15 mg, depending on the poisoning situation. 3 For anemia, take 15 mg orally daily. 4 When used in combination with 5-fluorouracil, take 20-30 mg/m2 orally, half an hour before taking 5-fluorouracil.
Adverse Reactions
Rarely, allergic reactions such as rash, hives, or asthma may occur occasionally.
Precautions
It is contraindicated in patients with pernicious anemia or other megaloblastic anemias due to vitamin B12 deficiency.

Drug Interactions
