Glipizide Tablets

Function and Efficacy

This product is a second-generation sulfonylurea antidiabetic drug. It is effective for most patients with type 2 diabetes, and can reduce fasting and postprandial blood sugar, and reduce glycosylated hemoglobin (HbAlc) by 1% to 2%. The main function of this type of drug is to stimulate pancreatic b cells to secrete insulin, but the prerequisite is that pancreatic b cells still have a certain function of synthesizing and secreting insulin. Its mechanism is to specifically bind to the sulfonylurea receptor on the b cell membrane, thereby closing the K channel, causing changes in membrane potential, opening the Ca2 channel, and increasing Ca2 in the cytosol, which promotes insulin secretion. In addition, there are extra-pancreatic effects, including improving the insulin resistance of peripheral tissues (such as liver, muscle, and fat).

Ingredients

Appearance

This product is white tablet or coated tablet.

Indication

It is suitable for patients with mild and moderate type 2 diabetes who have not achieved satisfactory results after 2 to 3 months of diet control and physical exercise. The pancreatic islet b cells of such diabetic patients must have a certain insulin secretion function, and have no acute complications (such as infection, trauma, ketoacidosis, hyperosmolar coma, etc.), no pregnancy, and no serious chronic complications.

Usage and Dosage

The oral dosage varies from person to person. The generally recommended dosage is 2.5 to 20 mg/day, taken 30 minutes before breakfast. If the daily dosage exceeds 15 mg, it should be taken three times before meals in the morning, afternoon and evening. For those who have failed to use diet therapy alone: the initial dosage is 2.5 to 5 mg per day, and the dosage is increased or decreased according to the blood sugar and urine sugar situation, increasing or decreasing by 2.5 to 5.0 mg each time. If the daily dosage exceeds 15 mg, it should be taken 2 to 3 times before meals. For those who have used other oral sulfonylurea hypoglycemic drugs: stop taking other sulfonylurea drugs for 3 days, and start taking this product after rechecking blood sugar. Gradually increase the dosage from 5 mg until the ideal therapeutic effect is achieved. The maximum daily dose does not exceed 30 mg.

Adverse Reactions

(1) The most common symptoms are gastrointestinal symptoms (such as nausea, upper abdominal distension) and headache, which can be relieved by reducing the dose. (2) Some patients may experience skin allergies. (3) Hypoglycemia may occur occasionally, especially in the elderly and weak, those who are overactive, those who eat irregularly, drink alcohol or have liver dysfunction. Use with caution.

Precautions

(1) Those who are allergic to sulfonamides. (2) Patients with confirmed diagnosis of type 1 diabetes. (3) Patients with type 2 diabetes accompanied by stress conditions such as ketoacidosis, coma, severe burns, infection, trauma, and major surgery. (4) Those with impaired liver and kidney function.

Special Population Medication

Precautions for children: Not tested. Precautions for pregnancy and lactation: 1. Animal experiments and clinical observations have shown that sulfonylurea hypoglycemic drugs can cause stillbirth and fetal malformation, so they are contraindicated for pregnant women. 2. This type of drug can be excreted through breast milk, so it is not suitable for nursing mothers to use it to avoid hypoglycemia in infants. Precautions for the elderly: Start with a small dose and gradually adjust the dose.
