Function and Efficacy

The pharmacological action of zoledronic acid is mainly to inhibit bone resorption. Its mechanism of action is not fully understood and may be related to multiple effects. Zoledronic acid can inhibit osteoclast activity in vitro, induce osteoclast apoptosis, and block osteoclast absorption of mineralized bone and cartilage by binding to bone. Zoledronic acid can also inhibit the enhanced osteoclast activity and bone calcium release caused by various stimulating factors released by tumors.

Appearance

This product is white or off-white loose mass or powder.

Indication

Hypercalcemia drugs, treatment of tumor bone metastasis and prevention of skeletal complications.

Usage and Dosage

Intravenous drip: For adults, 4 mg each time, diluted with 100 ml 0.9% sodium chloride injection or 5% glucose injection, then intravenously dripped. The drip time should be no less than 15 minutes. The drug should be administered once every 3 to 4 weeks or as directed by a doctor.

Adverse Reactions

The most common adverse reaction of this product is fever. Other adverse reactions mainly include: fatigue, chest pain, leg edema, conjunctivitis, nausea, vomiting, constipation, diarrhea, abdominal pain, dysphagia, anorexia, hypotension, anemia, hypokalemia, hypomagnesemia, hypophosphatemia, hypocalcemia, granulocytopenia, thrombocytopenia, pancytopenia, bone pain, joint pain, muscle pain; kidney: increased serum creatinine value, etc. (related to the time of administration); the toxic and side effects of zoledronic acid are mostly mild and transient. In most cases, they will automatically disappear within 24 to 48 hours without special treatment.

Precautions

Patients who are allergic to this product and other bisphosphonates are contraindicated. It is not recommended for patients with severe renal insufficiency.

Drug Interactions

This product should be used with caution when used in combination with aminoglycosides, because aminoglycosides have a synergistic effect in lowering blood calcium and may prolong the duration of hypocalcemia; when used in combination with diuretics, it may increase the risk of hypocalcemia; when used in combination with thalidomide, it will increase the risk of renal dysfunction in patients with multiple myeloma.
