Function and Efficacy

This product is a derivative of isonicotinic acid. Its mechanism of action is unknown. It may have an inhibitory effect on peptide synthesis. The effect of this product on Mycobacterium tuberculosis depends on the drug concentration at the infected site. It only has an antibacterial effect at low concentrations and a bactericidal effect at high concentrations. It inhibits the synthesis of mycolic acid in Mycobacterium tuberculosis. This product has partial cross-resistance with ethionamide.

Appearance

This product is an enteric-coated tablet, which appears yellow after removing the coating.

Indication

This product is only effective against mycobacteria. It is used in combination with other anti-tuberculosis drugs for patients with tuberculosis who have not responded to first-line treatment with drugs (such as streptomycin, isoniazid, rifampicin and ethambutol).

Usage and Dosage

1Used oral dose for adults, used in combination with other anti-tuberculosis drugs, 250 mg at a time, 2 to 3 times a day. 2Used oral dose for children, used in combination with other anti-tuberculosis drugs, 4 to 5 mg/kg per day, 3 times a day.

Adverse Reactions

1. The incidence rates are relatively high: depression (central nervous system toxicity). 2. The incidence rates are relatively low: unsteady gait or numbness, tingling, burning sensation, pain in hands and feet (peripheral neuritis), mental confusion or other mental changes (central nervous system toxicity), yellowing of eyes or skin (jaundice, hepatitis). 3. The incidence rates are extremely low: blurred vision or decreased vision, with or without eye pain (optic neuritis), menstrual disorders or fear of cold, decreased libido (male), dry and rough skin, hypothyroidism, joint pain, stiffness and swelling. 4. Pay attention if the following conditions persist: diarrhea, increased salivation, drooling, loss of appetite, metallic taste in the mouth, nausea, mouth pain, stomach pain, stomach discomfort, vomiting (gastrointestinal disorders, central nervous system toxicity), dizziness (including when getting up from a lying or sitting position), drowsiness, weakness (central nervous system toxicity).

Precautions

Pregnant women should not take it; children under 12 years old should not take it.

Special Population Medication

Precautions for children: Not suitable for children under 12 years old. Precautions for pregnancy and lactation: Pregnant women should not take. Precautions for the elderly: Not clear yet.

Drug Interactions

1. Taking it together with cycloserine may increase the incidence of central nervous system reactions, especially systemic convulsions. The dose should be adjusted appropriately and the central nervous system toxicity symptoms should be closely monitored. 2. The combination of this product with other anti-tuberculosis drugs may aggravate its adverse reactions. 3. This product is a vitamin B6 antagonist and can increase its renal excretion. Therefore, the requirement for vitamin B6 may increase in patients receiving prothionamide treatment.
