Function and Efficacy

This product is rapidly converted into the antiviral compound penciclovir in the body, which has an inhibitory effect on herpes simplex virus type Ⅰ (HSV-1), herpes simplex virus type Ⅱ (HSV-2) and varicella zoster herpes simplex virus (VZV). The mechanism of action is as follows: In cells infected with the above viruses, viral thymidine kinase phosphorylates penciclovir into monophosphate penciclovir, which is then converted into triphosphate penciclovir by cellular kinase. In vitro studies have shown that penciclovir triphosphate inhibits the activity of HSV-2 polymerase by competing with guanosine triphosphate, thereby selectively inhibiting the synthesis and replication of herpes virus DNA. In cell culture studies, penciclovir has an inhibitory effect on the following viruses (arranged in order of strength): HSV-1, HSV-2, VZV. The sensitivity of viruses to penciclovir is related to many factors. The qualitative change of viral thymidine kinase or DNA polymerase can lead to the production of HSV or VZV resistant mutants to penciclovir. It is common to see that mutants that are resistant to acyclovir due to the lack of viral thymidine kinase are also resistant to penciclovir. If the patient's clinical treatment is not effective, it should be considered that the virus may be resistant to penciclovir.

Appearance

This product is white tablets.

Indication

For the treatment of herpes zoster and primary genital herpes.

Usage and Dosage

This product is a dispersible tablet, which can be taken orally/swallowed directly, or put this product into 100ml of water, shake and disperse it, and then take it orally. Adults take 0.25g each time, 3 times a day, for 7 consecutive days. Patients with renal insufficiency should adjust the usage and dosage according to their renal function. The recommended dose is as follows: 0.25g for adults with creatinine clearance ≥60ml/min, 0.25g for adults with 40~59ml/min every 8 hours, 0.25g for adults with 20~39ml/min every 12 hours, 0.25g for adults with 20ml/min every 24 hours, 0.125g for adults with 20ml/min every 48 hours. Patients with compensated liver disease do not need to adjust the dose, and pharmacokinetic studies have not been conducted on patients with decompensated liver disease.

Adverse Reactions

1. Nervous system: dizziness, insomnia, drowsiness, abnormal sensation, etc.; 2. Digestive system: diarrhea, abdominal pain, indigestion, anorexia, vomiting, constipation, bloating, etc.; 3. Precautions

Contraindicated in patients allergic to famciclovir.

Special Population Medication

Precautions for children: The safety and efficacy of famciclovir for children have yet to be determined, so it is not recommended for children. Precautions for pregnancy and lactation: Pregnant and lactating women are generally not recommended to use this product. If this product is used, it should be confirmed that the benefits of using this product to the fetus or infant outweigh the risks. Precautions for the elderly: Unless there is renal insufficiency, the dosage can remain unchanged.
