Function and Efficacy

When it enters the human body, the ring structure of the sulfonate group opens and destroys the structure and function of DNA by alkylating guanine in the cell's DNA. The cytotoxic effect of this product is almost entirely manifested in the inhibition of hematopoietic function, mainly manifested in the obvious inhibition of granulocyte production. The second is the inhibition of platelets and red blood cells, and the inhibition of lymphocytes is very weak.

Indication

It is mainly used in the chronic phase of chronic myeloid leukemia, and has poor effects on patients lacking the Philadelphia chromosome Ph1. It can also be used to treat chronic myeloproliferative diseases such as essential thrombocythemia and polycythemia vera.

Usage and Dosage

Oral: Daily dose for adults 2-8 mg, children 0.05 mg per kilogram of body weight. Stop the drug or change to a maintenance dose (0.5-2 mg, once a day or twice a week) until the white blood cell count drops to 10,000-20,000. The efficacy of intermittent high-dose treatment of chronic myeloid leukemia is basically the same as the above method, but it can shorten the time of induction remission and may reduce the chance of aplastic anemia. Usage: According to the patient's white blood cell and platelet counts, three different doses of 80-100 mg, 50 mg, and 20-30 mg are selected for one administration (if 100 mg is taken at one time, it is generally taken in two doses and completed within 1 to 2 days). The interval between two doses is 1 to 2 weeks (depending on the blood picture). If 80-100 mg is taken at one time, it needs to be taken at least 2 weeks apart.

Adverse Reactions

Bone marrow suppression may occur. Common symptoms include granulocytopenia and thrombocytopenia. In severe cases, the drug should be discontinued immediately. Long-term use or excessive use may cause pulmonary fibrosis. Skin pigmentation, hyperuricemia and sexual dysfunction, feminization of male breasts, testicular atrophy, and irregular menstruation in women may occur. Cataracts, erythema multiforme rash, and polyarteritis nodosa are rare adverse reactions. There have been individual reports of epileptic seizures after high doses; endocardial fibrosis, and the corresponding symptoms; and rare hepatic venous atresia.

Precautions

This product may increase the risk of fetal death and congenital malformations, so it should not be used in the first three months of pregnancy. Patients who have been allergic to this drug in the past.
