What Is SS-31?

Elamipretide (SS-31) is a small peptide that can reach inside the mitochondria and affect mitochondrial function maintenance. Elamipretide belongs to a family of cell-penetrating peptides that target and accumulate at the inner mitochondrial membrane. Inside the mitochondria, elamipretide binds to cardiolipin, an essential component of the inner mitochondrial membrane, and restores the mitochondria. Given the effects of elamipretide on mitochondrial function and maintenance, treatment with elamipretide is under investigation to determine its benefits on human health. FUNCTIO N:

SS-31 has therapeutic effects on diseases related to free radicals, such as ischemia-reperfusion injury, neurodegenerative diseases, heart failure, and muscle aging. SS-31 is a mitochondrial-targeting molecule. It interacts with cardiolipin to reactivate ischemic mitochondria. At the same time, SS-31 is also a cell-penetrating peptide.

SS-31 is a small molecule composed of four amino acids. In vivo, SS-31 targets the inner membrane of mitochondria and can prevent oxidative damage to neurons and other cell types; prevent mitochondrial depolarization, reduce islet cell apoptosis, increase islet cell yield, and improve post-transplantation function.

A cell-permeable, synthetic tetrapeptide (D-Arg-Dmt-Lys-Phe-NH₂) that selectively concentrates in the inner mitochondrial membrane and reversibly binds to cardiolipin, a phospholipid essential for electron transport chain (ETC) function. By stabilizing cardiolipin, SS-31 restores mitochondrial bioenergetics, reduces cytochrome c peroxidase activity, and prevents oxidative damage in cells with impaired mitochondrial function.

Activity note:

SS-31 does not act via traditional receptor agonism. It binds cardiolipin with high selectivity in the inner mitochondrial membrane. Functional EC₅₀ values are context-dependent (cell type, stress model). Key electrophysiology studies report mitochondrial membrane potential restoration at nanomolar concentrations.
