The mechanism of action Tadestanol is a second-generation α5 inhibitor of 5-alpha reductase approved by the United States, and is used to treat benign prostatic hyperplasia. Tadestanol can inhibit the conversion of testosterone to dihydrotestosterone, which is an important component in the formation and maintenance of benign prostatic hyperplasia. During the treatment with tadestanol, the level of dihydrotestosterone decreases, but the serum testosterone level remains unchanged. Unlike α1-adrenergic receptor antagonists that take effect immediately, tadestanol takes several months to be effective for prostate size and symptoms of benign prostatic hyperplasia (hesitancy and difficulty in urination). Indications Tadestanol can be used to treat moderate to severe symptoms of Chemicalbook benign prostatic hyperplasia (BPH). For patients with moderate to severe symptoms of benign prostatic hyperplasia, it reduces the risk of acute urinary retention (AUR) and surgery. Drug interactions Tadestanol is metabolized by CYP3A4 in the liver. In vitro experiments show that CYP3A4 inhibitors such as ritonavir, ketoconazole, verapamil, diltiazem, cimetidine, and ciprofloxacin can increase the concentration of tadestanol in plasma, while there are no pharmacological or pharmacokinetic interactions with terazosin, terazosin and colestyramine.
