    1. Description

    Calycosin-7-O-glucoside is a natural isoflavone glycoside isolated from the roots of Astragalus membranaceus. Its molecular formula is C₂₂H₂₂O₁₀, and its molecular weight is approximately 446.40 Da. It appears as a white to off-white crystalline powder. Regarding solubility, it is soluble in DMSO (solubility exceeds 100 mg/mL) and slightly soluble in organic solvents such as methanol and ethanol. It should be stored at -20°C in a sealed container protected from light. The powder form can be stored for 2–3 years; once dissolved in a solvent, it is recommended to store the solution at -80°C and use it within 6 months. This compound exhibits various biological activities, including neuroprotective, cardioprotective, anti-inflammatory, and antioxidant effects.

    2. Application

    (1) Ischemic stroke research: In animal models of middle cerebral artery occlusion/reperfusion (MCAO/R), it is used to study the reduction of cerebral infarct volume, improvement of neurological deficits, and protection of blood-brain barrier integrity.

    (2) Neuroprotection Research: In oxygen-glucose deprivation/reoxygenation (OGD/R)-induced neuronal damage models, it is used to investigate the mechanisms underlying the reduction of oxidative stress, inhibition of apoptosis, and protection of neuronal survival.

    (3) Cardiovascular Disease Research: It is used to investigate the mechanisms of myocardial protection, exerting cardioprotective effects by regulating relevant signaling pathways.

    (4) Anti-inflammatory and Antioxidant Research: As a natural antioxidant, it is used to investigate the molecular mechanisms underlying the inhibition of inflammatory cytokine expression, the scavenging of free radicals, and the alleviation of oxidative damage.

    (5) Melanin Synthesis Research: In melanocyte models, it is used to investigate its effects on inhibiting tyrosinase activity and melanin biosynthesis.

    3. Main Efficacy

    (1) Neuroprotective effects: By activating the SIRT1/FOXO1/PGC-1α signaling pathway, it alleviates neuronal oxidative stress and apoptosis induced by oxygen-glucose deprivation and reoxygenation; in vitro experiments showed that treatment with a concentration of 40 μM increased the survival rate of HT22 hippocampal neurons by approximately 35% and reduced the percentage of TUNEL-positive cells by approximately 40%. In a cerebral ischemia-reperfusion injury model, a dose of 20 mg/kg reduced the infarct volume by approximately 25–45% and improved neurological function scores.

    (2) Protection of the blood-brain barrier: By regulating the NO/Cav-1/MMPs pathway, it inhibits the expression and activity of matrix metalloproteinases (MMP-2/MMP-9), protects tight junction proteins ZO-1 and occludin, and reduces blood-brain barrier permeability following cerebral ischemia. In a rat MCAO model, treatment reduced Evans Blue leakage by approximately 30–50%.

    (3) Antioxidant Effects: Concentration-dependently reduces the accumulation of reactive oxygen species (ROS) and malondialdehyde (MDA) levels within cells, while simultaneously increasing the activity of superoxide dismutase (SOD) and glutathione (GSH), thereby enhancing the endogenous antioxidant defense system.

    (4) Anti-apoptotic effects: Downregulates the expression of pro-apoptotic protein Bax, upregulates the expression of anti-apoptotic protein Bcl-2, and inhibits caspase-3/9 activity, thereby reducing neuronal apoptosis.

    (5) Anti-inflammatory effects: Inhibits the production and release of inflammatory cytokines, alleviating tissue damage caused by inflammatory responses.

    (6) Inhibition of melanin synthesis: By inhibiting tyrosinase activity (IC₅₀ value of 68 μM), it reduces melanin content; the 50% inhibitory concentration in Melan-a melanocytes is approximately 75.78 μM.

    (7) Cardioprotective effects: Exerts cardiomyoprotective effects through multiple signaling pathways, thereby reducing damage to cardiac tissue.
