Ky19382 for Hair Loss

KY19382, a novel indirubin analogue, has emerged as a promising candidate for hair loss treatment by targeting the Wnt/β-catenin signaling pathway, a key regulator of hair follicle development and cycling.

In preclinical studies, it acts as a dual inhibitor, blocking the interaction between CXXC5 and Dishevelled (Dvl) while inhibiting Glycogen Synthase Kinase 3β (GSK-3β) activity. This dual action effectively activates Wnt/β-catenin signaling, which in turn boosts the expression of alkaline phosphatase (ALP) and proliferation marker PCNA in dermal papilla cells—critical for maintaining hair follicle function.

Ex vivo experiments on mouse vibrissa follicles showed it significantly increased hair length. In vivo tests on mice further demonstrated its ability to induce hair regrowth and promote de novo hair follicle formation through wound-induced hair follicle neogenesis and hair patch assays. These findings position KY19382 as a potential therapeutic agent for alopecia, offering both hair regrowth and follicle regeneration benefits.
