1. The best condition for the EDC reaction is pH 4.5 (or between 4-6), and the efficiency is greatly reduced under neutral conditions.
2. The reaction time is too short, and it will be much better extended to 12h.
3. room temperature is enough.
4. After reaction, consider using dialysis or pillars to remove the small molecular weight.
