Distorted bands on Phos-tag gels-What's wrong?

Chen Tang  Follow

Hi all,

I think I found the answer: rotten MOPS.
I did my MOPS running buffer with the reagent available in the lab. This is a fairly old flask but the solutions were colourless (MOPS has a tendency to turn yellow when autoclaved) and, as said before, they were correct for use with no phos-tag gels. Only with phos-tag it was producing wrong patterns.
Today I changed the running buffer for a MES-based one and compared gels with and without Phos-tag. This time both gels run OK, while the Phos-tag one showed some band retention, as expected.
I hope it helps someone.

Cheers
