I assume you’re tritylating the amine, unless my brain isn’t working correctly. Your starting material may be water soluble but there’s almost no way Fmoc-Trityl-lysine prefers water over organic.More

Aqueous workup with 5% LiCl can help a lot too to remove the last bits of DMF; extract with chloroform, chloroform/isopropanol, or EtOAc. But I recommend speedvac most.More

Just to be clear, for extraction, I would use the the aqueous layer with EtOAc. Then I would rotovap the organic layer?More

I add some toluene to the flask and remove with the rotovap. The toluene forms a low boiling azeotrope with DMF. Edit: just saw that you tried that.More
