-There is no problem in washing bacterial cells/colonies in saline.0.89% Saline (NaCl) is used for washing bacterial cells for direct using them as cell templates. Saline washing helps to reduce high EPS (Exo-polysacharides)that may hinder cell lysis in PCR. Kindly see the following PDF for method of bacterial cell washing with saline (page 229-230) Molecular fingerprinting). Bacterial cells can be maintained in saline 0.89% for 2-3 months for using them as cell templates for amplification of genes using PCR.

Sodium chloride, or salt, is a nutrient that affects different organisms in different ways. For example, some organisms are obligate halophiles, which means they require salt to survive and will lyse, or break open, if the salt level drops to a low level. Other halophilic organisms are merely halotolerant, which means they don’t need salt to survive but can tolerate moderately salty environments. Halophiles belong to a larger group called extremophiles who thrive under extreme conditions. There may be some effect on bacteria.

Iam talking about normal bacteria, not tolerate bacteria . My question is can I use normal saline to wish bacteria and not effect on it.
