I have been priming my cells with PMA (20ng/ml) and Ionomycin (1ug/ml) for 24 hours, before 6 hours stimulation with different concentrations of IL-23, and I cant seem to get a big enough signal. We use 10 ng/ml in basolateral medium in a co -culture system -6 h preinduction then treatment with our samples and in parallel with our treatment, IL-6 stimulation continues till 10 hours total. We collect the the supernatant after 24 hours. I guess you can't find especificly a paper on IL-23 stimulation dosage. But, you have to test different doses to find the model has been established.
