Hi Christina, You should first determine the lethal concentration of G418 on un-transfected Jurkat cells. Plate your cells in multiwell plate and add your antibiotic at increasing concentrations. withing few days you would be able to determine the minimal lethal concentration for your cells. use this concentration for selection of your transfected cells. Good luck
