Ivy Li The solubility of hydrocortisone in methanol is around 6.2 mg/mL, much less than its solubility in DMSO (20 mg/mL), ethanol (15 mg/mL) and in DMF (30 mg/mL). If you stock solution of 5mg/mL or less works for you, methanol can be used.

Thanks for the reply Ejaj K Pathan . I don't need very high concentration and so methanol is fine. I'd like to know will or how these different solvents affect ELISA assay? Thank you.

The 10mg/mL stock can be prepared in 4% DMSO solution. This concentration of DMSO did not show any effect during the assay.
