You should cut at 5-6 micrometers and have a known positive control. The stain positivity is brick red, although you need a polarizing right to see the "apple green" stain. Not all types of amyloid stain and IHC should be preferred over the histochemical stain. Something can always go wrong if you do not have experience in histology and make sure you are using fresh solutions when performing the stain. I hope it helps.
