Do anyone Know the Superblock Blocking buffer composition?

Good Morning, the compositions is unknown. The specification doc from the website mention " The protein-based formulation does not contain any immunoglobulins, albumin, or endogenous biotin, making it compatible in many situations where traditional blocking agents fail."

In the past I run a SDS-PAGE gel to have an idea of whats inside and I saw multiple prroteins. The ideal would be to do Mass-spec of those bands but is out of the scope of most of us. Size exclusion also show multiple peaks and something that elute very slow that could be Tween?. Hope it helps.

Hi Balachandar , here i provide you the recipe we used.
2%  albumin,Bovine(BSA), 0.5% casein hydrolysate, 0.1% Tween-20 in PBS. and final Sterilize the solution by filtering it through a 0.22-um Millipore filter and keep it frozen at -20°C.

Thanks for your answer. I know 2% BSA and 2% SDS is efficient blocking buffer for ELISA. But I want Know the Super block Composition.

Hi Balachandrar, super block composition is a proprietary thing but you can try with 100-1000 mM of salt concentration   and 1-5 mM of chelating agents with the above composition ( By Huan).

10mM PBS+ 2% BSA will be efficient blocker for ELISA.
