How can i prepare different concentrations (0.5, 0.25, 0.1, 0.025,...
Hi there, Your question is not very clear about the volume you actually need for each concentration... Nevermind, the more concentrated solution is 0.5% so if you have in hand 0.5g you have enough to prepare 100mL of it. You then may generate the other solutions by simple dilution (0.25% is 2x dilution, 0.1% is 5x dilution aso.).

Dear Nouman You can first prepare 0.5% solution (0.5 g per 100 ml) and for 0.25, 0.1, 0.025 and 0.05%, respectively, dilute to half, one fifth, one twentieth and one tenth. For example, add 9 ml water to 1 ml of the stock solution, to 10 times dilute and obtain a concentration of 0.05%.

You can make only 100ml of 0.5% solution of compound or 0.25% 200 ml solution or 0.1% 500 ml solution consider reduction in 0.5% solution stock solution which may be 100 ml initially then dilute further

By keeping the solution volume constant : 250 ml/5= 50 ml, you can do it as follows : 0.5%= 0.25g/50ml, 0.25%=0.125g/50ml, 0.1%= 0.05g/50ml, 0.025%= 0.0125g/50ml, 0.05% = 0.025g/50ml, 0.25 + 0.125 + 0.05 + 0.0125 + 0.025 = 0.4625 g < 0.5 g.

first you should tell that, for how much volume you need to prepared the different concentration from stock and already you told in you first concentration is about 0.5% and you do this accordingly.

I think the best answer as Seied Mehdi Miri said firstly prepare a 0.5% solution by dissolving 0.5 g of colchicine in 100 ml of water. also to prepare a 0.25% solution you can take 50 ml from the solution 0.5% and dilute it to 100 ml by adding 50 ml of water. and to prepare a 0.1% solution you can take 20 ml of the 0.5 % solution and dilute it to 100 ml by adding 80 ml of water. also to prepare a 0.025% solution you can take 5 ml of the 0.5% solution and dilute it to 100 ml by adding 95 ml of water. and so on

prepare the highest concentration you need in large quantity and with the aid of the serial dilution formula you could prepare the other concentrations by picking from the highest concentration and dilution to get the other concentrations

how to prepare a stock solution of commercially available essential oil for checking invitro acaricidal activity , and how to make working solutions from that stock solution. plz guide . what is most common percentage of essential oil stock solution we use in acaricidal activity test. can we use commercially available oil ?

By keeping the solution volume constant : 250 ml/5= 50 ml, you can do it as follows :
0.5%= 0.25g/50ml,
0.25%=0.125g/50ml,

0.025%= 0.0125g/50ml,
0.05% = 0.025g/50ml,
0.25 + 0.125 + 0.05 + 0.0125 + 0.025 = 0.4625 g < 0.5 g.

What you should do is redo your calculations based on the fact that you only have enough to make 100 ml of a 0.5% solution.
Or buy more colchicine.
