Erman Kocak Thank you for your comments, If I could ask another adding question pertaining to your reply then do you know any other detergent that replaces SDS? Or is Native Page a better option in that case?

I think SDS-PAGE is inevitable in case of protein purification at least for the screening of purity of the samples that‘s why I would suggest you to use another buffer except potassium phosphate. Of course you can separate your samples with other electrophoresis methods which is not as routinized as SDS-PAGE for example with a cationic detergent;16-BAC based electrophoresis, but you should consider the preparation of 16-BAC gel that differs totally from SDS-Gels

Another commonly used buffer is Tris-HCl at pH 8.0. (Phosphate buffers can be adjusted into wider range of pH.) Tris has no interaction with SDS. SDS is an strong ionic detergent. NaOH and KOH are also stronger detergent and denaturation agents. Mild non-ionic detergents are different forms of Tween and Triton.

Both types of buffers may be used in many types of reactions. K-PO4 is the commonly used buffer and usually can be replaced with Na-PO4. The downstream reactions or interactions may wary slightly where potassium reacts with SDS. Your best bet is trying both buffers side-by-side and screening the final product to pick up the optimal one.
