You can perform wound healing assay to measure the rate of cell migration. I think the assay you are performing will result into cell invasion properties. However, I am not sure of latter. Please confirm it once again. Thanks!

Let me ask: how can a cell migrate if it doesn't have traction? Without attachment there is no traction and thus no migration. Ever tried to drive a car on wet ice? Same challenge - no traction, no motive force, no movement!

Hi, first of all if I am right Jurkat cells are precisely not supposed to migrate well, at least on a 2D substrate there is a striking contrast compared to primary T cells. A second thing is that the albumin in the FBS will limit cell adhesion , but from my experience with Jurkat cells, even in 10% FBS they will eventually form some adhesive contact with their substrate, although obviously not as strong as in the case of adherent cells.

Cells adhere to the chamber, nothing wrong! Probably, you have to put more cells and incubate longer time.
