Covalently closed circular DNA renatures readily once denaturing conditions are removed, either by cooling (heat denaturation) or neutralization (alkali denaturation); this is the principle of plasmid isolation. To give your biotinylated primers a chance to hybridize, you should add them together with the plasmid before proceeding to the denaturation-renaturation step
