Hi Xie, we prepare Cisplatin stocks in 1XPBS, and we aliquote them for like 20ul-50ul in each vial (amber microcentrifuge tubes) and keep these stocks at -20C.

Hi Ruth, thanks for your answer. It's ture about what you said. If the drug is dissolved in water, the acitivity of the drug will not be lost. However, the intact cisplatin in water will be lost, and this is a big question in bioanalysis. I still cannot find a good method for preparing the stock solution, it's really confused.

You have to dilute more. So that it would be dissolved. For example, if you want to make 5mM concentration, you may dissolve 3 mg cisplatin in 2ml 0.9% NaCl. I was succeeded.

It seems that many people have the same problem regarding the "correct" way of preparing the stock solution of cisplatin. I would like share my final solution to this problem. Based on my testing, the best way is to dissolve cisplatin in 0.9% NaCl, and this approach can keep cisplatin stable not only at room temperature but also at 37-41°C. The solubility of cisplatin in 0.9% NaCl is relatively low. The concentration of the stock solution is better less then 1 mg/ml (more preferably <=0.5 mg/ml). Moreover, the dissolution process is slow, you may need to wait for at least 2 h for a stock solution of 0.5 mg/ml to be completely dissolved.

Thanks Feifan Xie for sharing. I have another question how long did you store the stock solution. I have experiments that would run for 3 months was your stock solution stable for that long in 0.9% NaCl solution.
