What could happen when the Glycine is coupling in a sequence...
Hello! What can happen is that for (typically larger than 12 amino acids) longer sequences, peptide growth is compromised in relation to space. Thus, using the Fmoc strategy is not recommended for enabling the grouping (through hydrophobic interactions), and decreases the space (Fmoc group is large). In this sense, as glycine is a small amino acid (without side group), the protective Fmoc get closer to other groups, inducing agglomeration. For solution of this case, I recommend two alternatives: change to the t-Boc strategy and / or use the microwave synthesis (microwaves facilitate chain elongation and decrease intramolecular interactions).
