EDC and NHS activation is ofte performed at pH5-7  in MES buffer, and the reaction of an amine with the activated carboxylic acid works well in PBS at pH=7-8, so I would say in a one step process where both the -COOH and -NH2 are present,  just adding the EDC/HNS to a buffered solution close to physiological pH of the gelatin should work fine. Give it a go. If it is too slow, start off at slightly lower pH (say 6) and increase it after a few minutes of activation to 7.4 (before it crosslinks). Not sure what shape you want, but in stead of finding teflon-coatred moulds or coating moulds with Teflon, you could machine moulds directly out of Teflon stock.
