How lamda max. of trypan blue dye can be shifted from 580nm to 480nm?
Thank you Adam for your precious time. 1-In case of only Trypan blue dye, the major peak of maximum absorbance was around 580nm. However, in case of dye-NIPAM max. absorbance peak was shifted to 480 nm. Basically, I heated NIPAM-dye at 50 oC then cool down to room temperature. Then centrifuge it and use supernatant for checking absorbance. 2-Concentration of NIPAM microgel was very low and microgels particles might be settle down during centrifugation. 3- I did not control pH of this mixture.

Did you subtract the absorbance spectrum of dye-free NIPAM microgel? I would be concerned that light scattering by the microgel is making it appear that the absorbance of trypan blue shifted.

If you put free trypan blue through the same treatments as the trypan blue-NIPAM formulation, what happens to its absorbance spectrum?
