Function and Efficacy
Agomelatine is a 5-hydroxytryptamine 2C (S-HTx) receptor antagonist. Its antidepressant mechanism may be related to increasing the plasticity of neurons in the hippocampus and neuronal proliferation. Immunostaining was used to measure the proliferation, regeneration and death of neurons in the brain of adult rats. The results showed that long-term (3 weeks) administration of agomelatine can increase cell proliferation and neuronal regeneration in the ventral dentate gyrus of the hippocampus, which is related to emotional response. However, no similar situation was observed during acute or subacute administration (4 hours or 9 weeks). After continued prolonged administration, cell proliferation and neuronal regeneration occurred in the entire dentate gyrus area, indicating that agomelatine can increase neuronal regeneration in the hippocampus to varying degrees, thereby generating new granule cells. Patients with depression often have difficulty falling asleep, early awakening or changes in sleep rhythm. Polysomnography often shows a decrease in slow wave sleep (SWS), an increase in rapid eye movement sleep (REM) density or a decrease in latency, and a decrease in the proportion of delta sleep. Most antidepressants, such as tricyclic antidepressants (TCA) and selective 5-HT reuptake inhibitors (SSRI), have a regulating effect on REM, but have poor effects on non-REM sleep, especially SWS. Some drugs with 5-HT2 receptor blocking effects, such as mianserine and mirtazapine, have the effect of promoting sleep and improving sleep continuity, but their blocking effect can cause sleeplessness and daytime sleepiness. Agomelatine has a unique pharmacological mechanism, namely regulating the sleep-wake cycle, so it can regulate the patient's sleep structure at night and promote sleep.

Indication
The first melatonin receptor agonist with good antidepressant effect. Mainly used for antidepressant, anti-anxiety, sleep rhythm adjustment and biological clock regulation.
Usage and Dosage
Oral administration: 25 mg per day, taken at night. If the effect is not satisfactory after 2 weeks, increase the dosage to 50 mg (two tablets) per day. The course of treatment is 6 weeks. Or follow the doctor's advice.
Adverse Reactions
Common symptoms include headache, nausea, and fatigue. Gastrointestinal adverse reactions are rare.
