Valproamide suppository
Function and Efficacy
1 Pharmacology: This product is a broad-spectrum anticonvulsant drug of the valproic acid class. Its mechanism of action has not yet been elucidated. It may be through the inhibition of the γ-aminobutyric acid (GABA) degradation enzyme system in the brain to varying degrees, thereby increasing the concentration of GABA in the brain, while increasing the activity of glutamate dehydrogenase, thereby increasing the synthesis of GABA in the brain. On the other hand, preventing the reuptake of GABA by axons and glial cells can also increase the concentration of GABA in the synaptic cleft. Increase the postsynaptic inhibition achieved by GABA. Studies have shown that it can prevent the spread of abnormal discharges in epileptic lesions. 2 Toxicological animal experiments have shown that this product has no irritating effect on the rectal mucosa of rabbits administered rectal; experimental results and pathological examinations have shown that rabbits and dogs given large doses of valproic acid orally have no irritating effect on the rectal mucosa of rabbits.

Drug Interactions
1 This product is used in combination with general anesthetics or central nervous system depressants, the clinical effect of the latter may be more obvious. 2 When used in combination with anticoagulants such as warfarin or heparin, and thrombolytic drugs, the risk of bleeding increases. 3 When used in combination with aspirin or dipyridamole, it can prolong bleeding time due to reduced platelet aggregation. 4 When used in combination with phenytoin sodium and phenobarbital, the blood concentrations of phenytoin sodium and phenobarbital will increase. 5 When used in combination with primidone, it can also cause an increase in blood concentrations and lead to poisoning. If necessary, the dosage of primidone needs to be reduced. 6 When used in combination with clonazepam to prevent and treat absence seizures, there have been reports of a few cases where it induced absence seizures.
