Function and Efficacy

This product is a pyrimidine antimetabolite drug, which mainly inhibits dihydrofolate reductase, interferes with folic acid metabolism, and selectively inhibits the synthesis of DNA, RNA and protein. It is a cycle-specific drug and has no cross-resistance with alkylating agents.

Appearance

This product is white tablets.

Indication

This product is used for combined chemotherapy of ovarian cancer, SCLC, malignant lymphoma, and endometrial cancer, and is particularly effective for ovarian cancer and SCLC.

Usage and Dosage

Oral administration: 10-16 mg/kg per day, divided into four doses, 21 days as a course of treatment or 6-8 mg/kg per day, 90 days as a course of treatment. In the combined regimen, the recommended total dose is 150-200 mg/m2 per body surface area, for 14 consecutive days, with good tolerance. Taking it 1-1.5 hours after a meal or before bedtime can reduce gastrointestinal reactions.

Adverse Reactions

1. Severe nausea and vomiting are dose-limiting toxicities. Myelosuppression is mild to moderate, with a prominent leukocytopenia, which usually occurs one week after treatment and reaches the lowest point in 3 to 4 weeks. 2. Central or peripheral neurotoxicity occurs after long-term use and is a dose-limiting toxicity. It can be alleviated or disappear after stopping the drug for 4 to 5 months. 3. Occasionally, there may be hair loss, cystitis, rash, itching, weight loss, etc.

Precautions

It is contraindicated for those who are allergic to this product.

Special Population Medication

Precautions for children: Not yet clear. Precautions for pregnancy and lactation: Use this product with caution. Precautions for the elderly: Reduce the dosage for patients over 65 years old as appropriate.

Drug Interactions

Combination with monoamine oxidase inhibitors and antidepressants can cause severe orthostatic hypotension and should be used with caution. Combination with metoclopramide can cause dystonia. Simultaneous use with vitamin B6 may reduce peripheral neurotoxicity.
