Function and Efficacy

This product is a calcium influx blocker (also known as a calcium channel blocker or calcium ion antagonist), which can selectively inhibit the transmembrane influx of calcium ions into vascular smooth muscle and myocardial cells, especially for vascular smooth muscle. This product directly relaxes vascular smooth muscle and dilates peripheral arterioles, thereby reducing peripheral resistance (afterload) and significantly dilating coronary arteries. This product has a negative inotropic effect in the body, but has no effect on the in-body sinoatrial node and atrioventricular node.

Indication

It can be used alone or in combination with other antihypertensive drugs to treat hypertension.

Usage and Dosage

Oral administration: once a day, one tablet at a time, can be increased to once a day, two tablets at a time.

Adverse Reactions

The more common adverse reactions are headache, edema, fatigue, insomnia, nausea, abdominal pain, flushing, palpitations and dizziness. The less common adverse reactions are angina pectoris, hypotension, bradycardia, body odor hypotension, itching, rash, dyspnea, weakness, muscle spasm and indigestion. This product is similar to other calcium antagonists, with very few reports of adverse reactions such as myocardial infarction and chest pain, and these adverse reactions cannot be clearly distinguished from the patient's underlying diseases; no abnormal laboratory test parameters related to this product have been found.

Precautions

It is contraindicated in patients who are allergic to dihydropyridine calcium antagonists; it is contraindicated in the following situations: 1. Severe hypotension; 2. Aortic valve stenosis.

Special Population Medication

Precautions for children: There is no data on the use of this product in children. Precautions for pregnancy and lactation: The safety of this product for pregnant and lactating women has not been determined. In animal experiments, rats were given 10 mg/kg of this product. In addition to causing delayed delivery and delayed labor, intrauterine mortality increased by 5 times and the number of litters decreased by 50%. Therefore, this product can only be used when there are no other safer alternative drugs and the disease itself is more dangerous to the mother and child. There is no report on whether this product is excreted into breast milk. Precautions for the elderly: The peak time of this product's blood concentration is similar in elderly and young patients. The area under the drug-time curve (AUC) of elderly patients increases, the elimination half-life is prolonged, and the clearance rate has a downward trend. It has been reported that when receiving similar doses of amlodipine, elderly patients have the same good tolerance as young patients. Therefore, elderly patients can use normal doses, but it is better to start with a smaller dose and then gradually increase the dose.

Drug Interactions

1. Anesthetics: Inhaled hydrocarbon drugs can cause hypotension when used with this product. 2. Nonsteroidal anti-inflammatory drugs, especially indomethacin: Used with this product can weaken the antihypertensive effect, which may be related to the inhibition of prostaglandin synthesis and (or) causing water and sodium retention. 3. β-receptor blockers: Used with this product is well tolerated, but can cause excessive hypotension, and in rare cases can increase the possibility of congestive heart failure. 4. Estrogen: Used with this product can increase fluid retention and increase blood pressure. 5. Sulfinpyrazone: Used with this product can increase the protein binding rate of amlodipine and change the blood drug concentration. 6. Lithium preparations: Used with this product, it can cause neurotoxicity, manifested by nausea, vomiting, diarrhea, ataxia, tremor and (or) numbness, and must be used with caution. 7. Sympathomimetic amines can weaken the antihypertensive effect of this product.
