Melanotan(MT)-II is a potent, non-selective melanocortin receptor agonist with high affinity for MC1,MC3,MC4, and MC5 receptor subtypes involved in the regulation of many physiological systems, such as the pigment system, energy homeostasis, sexual function, immune system, inflammation, and cardiovascular system [1].
In vivo studies of melanotam (MT)-II exert dose-dependent induced activity on erections by triggering erectile events and shortening the latency of the first erectile event that occurs. After intravenous injection of melanotan(MT)-II(1 mg/kg), the erection response induced by cavernosic nerve stimulation increased, thus promoting erection [2]. Melanotan (MT)-II promotes peripheral nerve regeneration and has neuroprotective properties in rats. Melanotan (MT)-II significantly enhanced sensory function recovery after sciatic nerve compression lesions in rats at doses of 20μg/ kg/48h,sc, but not at doses of 2 or 50μg/ kg [3]. Melanotan(MT)-II is an effective initiator of penile erection in men with erectile dysfunction [4]. Melanotan(MT)-II reduces food intake and body weight and causes thermogenic responses in mouse models [5].
Animal experimental rats: To investigate the activity of the inducer on erectile erection, after obtaining a 5-minute baseline recording period, intravenous injection, acute injection of melanotan (MT) -II or the vector (saline) in or within PVN was given. ICP and MAP were then recorded for 60 minutes after delivery of either saline or Melanotan (MT) -II. Iv injection (three doses; 0.1,0.3 and 1mg/kg) in saline were administered with a catheter inserted into the jugular vein [2]. Mice: Given melanotan (MT) -II (0.1 and 0.2nM) or a carrier (artificial cerebrospinal fluid). The SPA records continuously every 5 minutes. Food intake was measured at 6 and 24 hours after injection. Weight is measured every 24 hours. Food and water are provided at will. There is a 48-hour interval between drug treatments [5].
