Trazodone is a triazole pyridine antidepressant with similar effects to tricyclic and MAOI. It can be clinically used to treat depression. For patients with refractory depression who have not been treated with other antidepressants, this product is often effective. It can also be used to treat anxiety disorders, and is more suitable for treating elderly patients with depression or accompanying heart diseases. The mechanism of action is to selectively block the reuptake of 5-HT and have a weak effect on blocking the reuptake of norepinephrine, but it has no effect on dopamine, histamine, and acetylcholine, nor does it inhibit the activity of MAO in the brain. It has low toxicity to the cardiovascular system and is more suitable for elderly or depression patients with cardiovascular disease. This product also has central sedative and slight muscle relaxation effects, but has no anti spastic or central excitatory effects. It has no effect on the Chemicalbook heart, but can cause a decrease in blood pressure and is dose-dependent. After oral administration, it can be well absorbed, and the peak plasma concentration occurs about 1 hour after taking trazodone hydrochloride on an empty stomach, and 2 hours after taking it together with food. After absorption, it is mostly distributed in the liver and kidneys, and through liver metabolism, the metabolites still have significant activity. Easy to pass through the blood cerebrospinal fluid barrier, but rarely through the placental barrier. Metabolites are ultimately excreted through the kidneys. Rarely excreted in its original form. Renal function damage does not affect its excretion. T1/2 is 4.1h. The elimination of trazodone hydrochloride occurs in two phases, including an initial phase (T1/2 for 6 hours) and a subsequent slow phase (T1/2 for 5-9 hours). Due to significant changes in the in vivo clearance rate of trazodone hydrochloride, some patients may accumulate in the plasma after taking it.
