    Overview
    S-1-amino-2-propanol is an intermediate for the synthesis of nucleoside drugs such as the anti-AIDS drug tenofovir, and also has important applications in the research and development of other new drugs and new materials.
    Synthesis method
    Step (1): Dissolve sodium tert-butoxide (230g, 2.39mol) in tetrahydrofuran (2L), slowly add trifluoroacetamide (226g, 2mol) in an ice bath, and stir for 30min; then add S-propylene oxide (128g, 2.2mol) in an ice bath, naturally warm to room temperature, continue stirring and reacting for 10h, and stir and react at 35℃ for another 2h; after the reaction, add 2N (equivalent concentration) hydrochloric acid in total 1L to the system for neutralization, then add 1L of water, stand for stratification, extract the aqueous phase with dichloromethane, combine the organic phases, dry over anhydrous sodium sulfate, and concentrate under reduced pressure to obtain 318g of intermediate product I with a yield of 93%. Step (2): The intermediate product I (310 g, 1.81 mol) obtained in step (1) was dissolved in methanol (1.5 L), and 200 mL of water and potassium carbonate (550 g, 4 mol) were added. After stirring at room temperature for 6 h, the mixture was filtered and concentrated to dryness.
