Upadacitinib has shown excellent efficacy in clinical trials for the treatment of various autoimmune diseases and inflammatory disorders. Upadacitinib is a small molecule oral targeted drug developed by the internationally leading biopharmaceutical company AbbVie. It was approved in 2019 for the treatment of atopic dermatitis (AD), rheumatoid arthritis (RA), and psoriatic arthritis (PsA) in adults and adolescents aged 12 and above, becoming the most comprehensive selective JAK1 inhibitor available in China at present.

In vitro study
In biochemical tests, avarotinib has a selectivity 74 times higher for JAK-1 than for JAK-2 (which is involved in red blood cell production), and 58 times higher for JAK-1 than for JAK-3 (which is related to immune surveillance). The enhancement of the selectivity of UAADACITIB for JAK-1 over JAK-2 and JAK-3 in the RA range may provide an improved risk-risk distribution.
In vivo study
The second JAK inhibitor developed by AChemicalbookbbVie, Upadacitinib, is described as a second-generation inhibitor. The multi-dose Phase I study was completed in 2013. In the Phase I trial, using an immediate-release formulation, it was found that Upadacitinib was safe and well-tolerated up to 24mg, twice daily. The exposure of Upadacitinib in the evaluated multi-dose was proportional to the dose.
