HistaDAO-P™ Animal-derived Diamine Oxidase: A Scientific and Effective Solution Diamine oxidase in the human body is a key enzyme that catalyzes the oxidative deamination of histamine, converting exogenous histamine in food into inactive metabolites, preventing its absorption into the bloodstream and causing histamine intolerance. However, when diamine oxidase activity is insufficient, even a normal amount of histamine intake can lead to symptoms. ❶ A double-blind, randomized, placebo-controlled trial (n=400) conducted by Sánchez-Pérez et al. showed that taking enteric-coated mini-tablets containing porcine or plant-derived diamine oxidase (4.2 mg/dose or 8.4 mg/dose) 20 minutes before meals (3 times daily), combined with a low-histamine diet or a Mediterranean diet, significantly improved gastrointestinal, skin, respiratory, and headache symptoms related to histamine intolerance (primary endpoint), with better results than the intervention with placebo combined with a regular diet.

❷ Maintz and Novak (2007), in a systematic review, pointed out that exogenous supplementation of diamine oxidase significantly improved dietary freedom for over 80% of patients with histamine intolerance (HIT), allowing them to safely consume foods containing moderate amounts of histamine. Simultaneously, patients' quality of life scores (covering improvements in gastrointestinal, skin, neurological, and other multi-system symptoms) showed statistically significant improvements, providing important evidence-based support for the use of diamine oxidase supplements in the symptom management of HIT patients.

Market Quality Deficiencies

There are serious deficiencies in market quality control. Analysis of commercially available diamine oxidase products shows significant variations in enzyme activity between products.

Inconsistent Activity

The deviation between measured activity and the labeled value ranges from 0% to 244.445%. In an evaluation of commercially available products, only two out of ten supplements analyzed had diamine oxidase activity that met or exceeded the labeled HDU content.

Porcine-derived diamine oxidase has the most mature research foundation, a clear biochemical identity, and reliable catalytic capacity. High-specification extraction and strict QC can mitigate the challenges of activity variation. Plant-derived diamine oxidase is less studied than porcine kidney diamine oxidase and performs poorly in commercial product testing. Of the three plant-derived products analyzed, only one detected identifiable diamine oxidase activity.
