Description	Tiletamine is a dissociative anesthetic and pharmacologically classified as an NMDA receptor antagonist. It is related chemically to ketamine. Tiletamine hydrochloride exists as odorless white crystals. It is used in veterinary medicine in the combination product Telazol (tiletamine/zolazepam, 50 mg/ml of each in 5 ml vial) as an injectable anesthetic for use in cats and dogs. It is sometimes used in combination with xylazine (Rompun) to chemically immobilize large mammals such as polar bears and wood bison. Telazol is the only commercially available tiletamine product in the United States. It is contraindicated in patients of an ASA score of III or greater and in animals with CNS signs, hyperthyroidism, cardiac disease, pancreatic or renal disease, pregnancy, glaucoma, or penetrating eye injuries.

Tiletamine (1-(2-thienyl)-2-methyl-2-aminopropan-1-one) is a dissociative anesthetic drug. It is chemically similar to ketamine and phencyclidine (PCP). While it possesses potent anesthetic properties, it is rarely used in its pure form. Instead, it is almost exclusively utilized in a combination formulation with the sedative zolazepam.

1. Medical Use

Tiletamine is primarily used in veterinary medicine. It is the active ingredient in the brand name drug Telazol.

Mechanism of Action: Like other dissociatives, tiletamine acts as an antagonist at the NMDA (N-methyl-D-aspartate) receptor. This blocks pain signals from reaching the brain, producing a trancelike state while the patient remains physiologically stable.

Veterinary Applications: It is used for the induction and maintenance of anesthesia in a wide range of animals, including cats, dogs, and exotic species. It is particularly valued for its rapid onset and strong muscle relaxation properties.

2. Recreational Use

Although designed for animals, tiletamine is sometimes diverted for human recreational use due to its psychoactive effects.

Effects: Users report effects similar to ketamine, including dissociation (a feeling of detachment from one's body), hallucinations, euphoria, and analgesia.

Risks: Recreational use of tiletamine is extremely dangerous. Because the drug is typically obtained from veterinary clinics, it is usually in the form of Telazol (a mix of tiletamine and zolazepam). The synthesis involves reacting 2-acetylthiophene with ammonium chloride and sodium cyanide to form an aminonitrile intermediate, which is then treated with methylmagnesium iodide to yield the final product.
