1. Description

Fusidic Acid is a high-purity functional raw material with mild and stable properties. 2. Application

(1) Treatment of skin and soft tissue infections: Used to treat skin infections caused by susceptible bacteria such as Staphylococcus, Streptococcus, and Propionibacterium acnes, including impetigo, boils, carbuncles, paronychia, wound infections, folliculitis, and acne vulgaris. Apply topically 2–3 times daily for a 7-day course of treatment.

(2) Treatment of bone and joint infections: Used for the treatment of deep staphylococcal infections such as osteomyelitis and suppurative arthritis, administered orally or intravenously. The standard adult dose is 500 mg three times daily.

(3) Treatment of systemic infections: Used for severe infections such as pneumonia and sepsis caused by methicillin-resistant Staphylococcus aureus (MRSA), often requiring combination with other anti-staphylococcal agents.

(4) Research on Mycoplasma genitalium infections: Recent studies indicate that fusidic acid exhibits good activity against Mycoplasma genitalium, with a minimum inhibitory concentration (MIC) range of 0.31–4 μg/mL, suggesting it may serve as a potential therapeutic option for drug-resistant Mycoplasma genitalium infections.

(5) Research tool compound: As a bacterial protein synthesis inhibitor, it is used to study ribosomal function, elongation factor mechanisms, and the development of bacterial resistance.

3. Main Efficacy

(1) Broad-spectrum activity against Gram-positive bacteria: Highly active against Staphylococcus spp. (including methicillin-resistant Staphylococcus aureus), and exhibits some antibacterial activity against Streptococcus spp., Corynebacterium diphtheriae, Clostridium spp., Neisseria spp., and Mycobacterium tuberculosis. Cross-resistance occurs with other antibiotics that lack target-specificity.

(2) Inhibition of bacterial protein synthesis: It binds to the elongation factor G-guanosine diphosphate complex, preventing its release from the ribosome and arresting protein synthesis, thereby exerting a bacteriostatic effect; at high concentrations, it exhibits bactericidal activity.

(3) Oral absorption and tissue distribution: It is well absorbed orally, with peak plasma concentrations reached 2–3 hours after administration on an empty stomach; plasma protein binding is as high as 97–99%. It is widely distributed in the body but does not readily cross the blood-brain barrier; it can enter the fetal circulation and breast milk.

(4) Safety of topical application: Topical formulations have low penetration rates under normal skin conditions but can penetrate deep into the skin and reach the site of infection under pathological conditions. Systemic absorption following topical application is minimal, and the incidence of adverse reactions is less than 1%.

(5) Hepatobiliary excretion characteristics: It is primarily metabolized in the liver and excreted via bile; approximately 100% is excreted in feces, with renal excretion accounting for less than 1%. The plasma half-life is 5–6 hours.
