Climbazole Targets Dandruff and Relieves Scalp Itchiness Effectively

1. Description

Climbazole is a synthetic imidazole antifungal compound with the chemical name 1-(4-chlorophenoxy)-1-(1H-imidazol-1-yl)-3,3-dimethylbutan-2-one. It appears as a white to off-white crystalline powder with a melting point of 96–100°C. Its molecular formula is C₁₅H₁₇ClN₂O₂, and its molecular weight is approximately 292.76. The compound is soluble in methanol, practically insoluble in water, and has a solubility of approximately 5 mg/mL in dimethyl sulfoxide (DMSO). As a chiral molecule, Ganboside exists as a racemic mixture, consisting of equal amounts of (R) and (S) enantiomers. Storage conditions: Store at room temperature in a cool, dry place protected from light (recommended <15°C). 2. Application

(1) Development of anti-dandruff products: As a broad-spectrum antifungal agent, it is widely added to hair care products such as anti-dandruff shampoos and conditioners for the treatment and prevention of dandruff. It works by inhibiting the growth of scalp-associated fungi, such as Malassezia. Brand names include Crinipan AD, among others.

(2) In vitro Antifungal Studies: Used to investigate the inhibitory activity of climbazole against pathogenic fungi such as Candida and Malassezia. Studies indicate that the minimum inhibitory concentration (MIC) of climbazole against Candida albicans is 0.29 μg/mL, and the MIC range against Malassezia glabrata is <0.06–1 μg/mL. Its activity is even higher than that of clotrimazole in certain strains.

(3) Cytochrome P450 enzyme studies: As a potent inducer and inhibitor of rat liver cytochrome P450, it is used to investigate the regulatory mechanisms of drug-metabolizing enzymes. Studies show that Ganbaoside increases cytochrome P450 levels in rat liver microsomes.

(4) Ecotoxicological studies: Used to investigate the fate and ecotoxicity of ganbocidin in the environment. This compound exhibits persistence in soil (DT₅₀ approximately 300 days) and exhibits moderate toxicity to fish and bees.

(5) Keratinocyte differentiation studies: Used to investigate the role of ganbocidin in promoting the expression of envelope proteins in keratinocytes.

3. Main Efficacy

(1) Inhibition of ergosterol biosynthesis: Ganbaoside interferes with the synthesis of ergosterol in the fungal cell membrane by specifically inhibiting cytochrome P450 14α-demethylase (lanosterol 14α-demethylase). Ergosterol is a key structural component of the fungal cell membrane; inhibition of its synthesis leads to increased cell membrane permeability, ultimately resulting in fungal cell death.

(2) Broad-spectrum antifungal activity: It exhibits inhibitory effects against pathogenic fungi such as Malassezia and Candida. In Malassezia furfur, the MIC range of climbazole is <0.06 to 1 μg/mL, with a median value of 0.06 μg/mL.

(3) Modulation of cytochrome P450 enzyme activity: climbazole can induce the expression of cytochrome P450 in rat livers. This effect may influence the metabolism of other drugs and should be considered in drug interaction studies.

(4) Increases expression of keratinocyte envelope proteins: In primary keratinocytes, ganbaoside increases the expression of keratinocyte envelope proteins; this effect may be related to its efficacy in improving scalp barrier function..
