    1. Description

5-Deazoflavin is a synthetic analog of riboflavin (vitamin B2) with a core structure of pyrido[4,5-b]quinoline-2,4(1H,3H)-dione, obtained by replacing the nitrogen atom at the N-5 position of the isoquinoleine ring with a carbon atom. This structural modification confers unique electrochemical and photochemical properties on the compound, making it a two-electron hydride carrier rather than a single-electron carrier, and endowing it with a low redox potential. It appears as a powder, with the molecular formula C₁₁H₇N₃O₂ and a molecular weight of approximately 213.2 Da. Storage conditions require sealed, light-protected storage at room temperature; during transportation, it must be kept sealed and protected from light at 2–8°C. This compound exists in nature as a precursor to coenzyme F₄₂₀ and 8-hydroxy-5-denitroflavone, playing a key role in microbial metabolism and the function of photolases.

    2. Application

    (1) Anticancer Drug Development: As an inhibitor of the HDM2-p53 interaction, these compounds are being investigated as potential therapeutic strategies for wild-type p53 tumors. 5-Deazaflavin derivatives stabilize the p53 protein and induce the expression of its downstream target genes by inhibiting the E3 ubiquitin ligase activity of HDM2. The novel derivatives exhibit nanomolar-level cytotoxic activity against MCF-7 breast cancer cells and HeLa cervical cancer cells.

    (2) Antimicrobial and Antituberculosis Research: As substrate analogues of F₄₂₀-dependent oxidoreductases, these compounds are used to investigate the metabolic pathways of actinomycetes such as Mycobacterium tuberculosis. F₄₂₀-dependent enzymes can activate the anti-tuberculosis prodrugs pretomanid and delamanid, making them of significant value in the treatment of tuberculosis.

    (3) Photolyase mechanism studies: As a structural analog of 8-hydroxy-5-deazaflavin, it is used to investigate the light-harvesting mechanism of DNA photolyases. Fo acts as a cofactor for photolyases in archaea, bacteria, and eukaryotes, participating in the repair of UV-induced pyrimidine dimers.

    (4) Environmental remediation research: Used to investigate the degradation mechanisms of F₄₂₀-dependent enzymes toward environmental pollutants, including the biotransformation of harmful substances such as picric acid, aflatoxins, and malachite green.

    (5) Antibiotic biosynthesis research: Used to study the biosynthetic pathways of tetracyclines and pyrrolo[3,4-b]indole antibiotics in actinomycetes such as Streptomyces; F₄₂₀-dependent oxidoreductases play a key role in the synthesis of these secondary metabolites.

    3. Main Efficacy

    (1) HDM2 E3 ligase inhibitory activity: 5-Deazaflavin derivatives selectively inhibit HDM2-mediated p53 ubiquitination, stabilize p53 protein levels, and activate p53-dependent apoptosis pathways. Structure-activity relationship studies indicate that a trifluoromethyl or chloro substituent at the C9 position, as well as additional halogen or methyl substituents on the benzene ring at the N10 position, significantly enhance inhibitory activity.

    (2) Antitumor cell proliferation activity: Novel 5-deazaflavin derivatives exhibit potent inhibitory effects on MCF-7 breast cancer cells and HeLa cervical cancer cells, with the half-maximal inhibitory concentration (IC₅₀) of some compounds reaching 0.1 nM. Kinase profiling analysis reveals that these compounds possess inhibitory activity against multiple protein tyrosine kinases.

    (3) Mimicry of F₄₂₀ Cofactor Function: As structural analogs of F₄₂₀, these compounds participate in low-potential two-electron redox reactions, serving as electron carriers in the methanogenesis pathway of methanogenic archaea and in various metabolic processes of actinomycetes.

    (4) DNA damage repair assistance: As an analog of the light-harvesting chromophore Fo in photolyases, it participates in the photoreactivation repair process of UV-induced DNA pyrimidine dimers.

    (5) Prodrug activation: In actinomycetes such as Mycobacterium tuberculosis, F₄₂₀-dependent nitro-reductases activate the anti-tuberculosis prodrugs pretomanid and delamanid, generating bactericidal active metabolites.

    (6) Biodegradation Promotion: F₄₂₀-dependent redox enzymes catalyze the reductive degradation of various environmental pollutants, including picric acid, aflatoxins, and malachite green.
