    1. Description

    Niacinamide is the amide form of nicotinic acid (vitamin B3) and belongs to the water-soluble B-complex vitamins. It appears as a white crystalline powder, is odorless or nearly odorless, and has a bitter taste. Its molecular formula is C₆H₆N₂O, and its molecular weight is approximately 122.12 Da. Its melting point is 128–131°C, and its boiling point is 150–160°C. The compound is highly soluble in water (approximately 100 mg/mL), ethanol, and glycerol, and is soluble in dimethyl sulfoxide; its aqueous solution is neutral. It should be stored in a light-protected, sealed container at room temperature or 2–8°C. As a precursor to nicotinamide adenine dinucleotide (NAD) and nicotinamide adenine dinucleotide phosphate (NADP), nicotinamide plays a key role in cellular energy metabolism, redox reactions, and DNA repair, and also acts as an inhibitor of poly(ADP-ribose) polymerase and sirtuin longevity proteins. This compound is classified by the U.S. Food and Drug Administration (FDA) as Generally Recognized as Safe (GRAS) and is widely used in the pharmaceutical, food, and cosmetics industries.

    2. Application

    (1) Skin Care and Cosmetics: As a multifunctional skincare ingredient, it is added to serums, creams, lotions, face masks, and other products to improve skin barrier function, reduce pigmentation, combat aging, control oil production, and minimize pores.

    (2) Clinical Medical Treatment: Used to prevent and treat niacin deficiency disorders such as pellagra, stomatitis, and glossitis. As a prescription medication (e.g., an active ingredient in Niaspan), it is used to lower total cholesterol, LDL cholesterol, and triglyceride levels in patients with hyperlipidemia.

    (3) Diabetes Research: In type 1 diabetes research, it is used to protect β-cell function and slow disease progression. In animal models of diabetic nephropathy, nicotinamide exerts a renoprotective effect by reducing oxidative stress and inflammatory responses.

    (4) Neurodegenerative Disease Research: Used in studies of neurodegenerative diseases such as Alzheimer’s disease and Parkinson’s disease to explore its neuroprotective potential through maintaining NAD⁺ levels and inhibiting excessive PARP activation.

    (5) Oncology Research: As a PARP inhibitor, it is used to investigate mechanisms for enhancing sensitivity to radiotherapy and chemotherapy, as well as its application in the prevention of certain types of skin cancer.

    (6) Anti-inflammatory and Renal Protection Research: It is used in studies of acute kidney injury models, such as renal ischemia-reperfusion injury and contrast-induced nephropathy, to mitigate kidney damage through anti-inflammatory and antioxidant mechanisms.

    3. Main Efficacy

    (1) NAD⁺ precursors and energy metabolism: As a precursor to NAD⁺, it participates in hundreds of redox reactions within cells and supports energy metabolism processes such as glycolysis, the citric acid cycle, and oxidative phosphorylation. Unlike nicotinic acid, nicotinamide does not cause flushing because it does not activate G protein-coupled receptors.

    (2) PARP Inhibition and Sirtuin Regulation: As a PARP inhibitor, it reduces DNA damage-induced cellular energy depletion; simultaneously, as a sirtuin inhibitor, it modulates deacetylase activity. Nicotinamide is an inhibitor of SIRT1 and SIRT2 (IC₅₀ for SIRT1 is 51.9 μM), rather than an activator.

    (3) Improvement of skin barrier function: Topical application increases the synthesis of ceramides and free fatty acids in the stratum corneum, thereby strengthening the skin barrier, reducing transepidermal water loss, and increasing skin hydration.

    (4) Inhibition of melanin transfer: By inhibiting the transfer of melanosomes from melanocytes to keratinocytes, it reduces melanin deposition in the epidermis, thereby brightening skin tone and lightening dark spots. This is the core mechanism of niacinamide’s skin-brightening effect.

    (5) Anti-inflammatory and Immune Modulation: Inhibits the NF-κB signaling pathway, reducing the production of pro-inflammatory cytokines such as interleukin-1β, interleukin-6, and tumor necrosis factor-α. In acne and rosacea, topical niacinamide can alleviate inflammatory responses, with efficacy comparable to 1% clindamycin gel.

    (6) Oil Control and Pore Minimization: By inhibiting sebum secretion from sebaceous glands, it reduces facial oiliness. Studies indicate that 2% niacinamide significantly reduces sebum secretion rates after 4 weeks of use, while simultaneously improving the appearance of enlarged pores.

    (7) Anti-Aging Effects: By promoting collagen synthesis, inhibiting matrix metalloproteinase activity, and protecting cells from oxidative stress damage, it slows down the skin aging process.

    (8) Renal Protection: Through anti-inflammatory and antioxidant mechanisms, it alleviates tubular damage and improves renal function in models of diabetic nephropathy and ischemia-reperfusion injury.

    (9) Neuroprotection: In models of neurodegenerative diseases, it protects neurons from damage by maintaining NAD⁺ levels, inhibiting excessive PARP activation, and reducing oxidative stress.

    An amine and amide. Acts as a weak base in solution. Amines are chemical bases. They neutralize acids to form salts plus water. These acid-base reactions are exothermic. The amount of heat that is evolved per mole of amine in a neutralization is largely independent of the strength of the amine as a base.
