A gastroprokinetic agent developed by Takeda. It appears as a solid white to off-white crystalline powder with a purity of ≥98% . It is soluble in DMSO (≥5 mg/mL) and should be stored at 2-8°C (refrigerated)
Mechanism of Action
It acts as a selective 5-HT4 receptor agonist and a 5-HT3 receptor antagonist . By stimulating 5-HT4 receptors in the gut, it facilitates the release of acetylcholine from enteric cholinergic neurons, which enhances gastrointestinal motility without affecting dopamine D2 receptors.
Applications
Therapeutic: Clinically used to treat upper digestive symptoms caused by chronic gastritis, including heartburn, nausea, and vomiting. It is also effective for functional dyspepsia and gastro-oesophageal reflux disease
Research: Used as a tool to study serotonin receptor activity in the digestive tract
Safety and Side Effects
Common Side Effects: Diarrhea, loose stools, thirst, and fatigue
Serious Risks: Rare cases of liver dysfunction (fulminant hepatitis, jaundice) have been reported. If appetite loss or yellowing of the skin occurs, medical attention is required
Toxicology: Non-genotoxic. The acceptable daily intake (ADI) established by the Food Safety Commission of Japan is 0.03 mg/kg bw/day
