CUCURBITACIN IIA

Cucurbitacin IIa (also known as Hemslecin A or Dihydrocucurbitacin Q1) is a tetracyclic triterpenoid primarily isolated from the root of Hemsleya amabilis . Like its close analog Cucurbitacin IIb, it serves as the main active component in approved anti-inflammatory prescription therapies (such as Hemslecin capsules and tablets) in China. 

• Actin Cytoskeleton Disruption: It causes the irreversible clustering and aggregation of filamentous actin (F-actin). This disrupts the cell’s structural integrity and halts mitosis.
• Cell Cycle Arrest & Apoptosis: It stalls the cell cycle at the G2/M phase. It drives PARP- and caspase-3-dependent cell death by downregulating survivin (an inhibitor of apoptosis protein).
• • EGFR Inhibition: It acts as a blood-brain barrier-permeable EGFR inhibitor (with an IC50 of 1.455 nM), making it highly effective in non-small cell lung cancer and colorectal cancer research.

  Cucurbitacin IIa functions as a non-steroidal anti-inflammatory component: 
  • Pathway Inhibition: It dampens systemic inflammation by blocking the EGFR-MAPK signaling pathway.
