Because cancer cells rely heavily on Hsp90 to keep unstable, mutated oncogenes folded correctly, 17-AAG effectively destabilizes the internal support system of a tumor.

1. Degradation of Oncogenic "Client Proteins"

Hsp90 acts as a protective shield for many critical signaling proteins. When 17-AAG blocks Hsp90, these unprotected "client proteins" are misfolded and immediately marked for destruction via the ubiquitin-proteasome pathway. Key client proteins degraded by 17-AAG include:
