    Introduction

    Creatine is naturally synthesized from amino acids in the human liver and transported to muscle cells via blood circulation, where it is converted into creatine salt. Human muscle movement is powered by the decomposition of adenosine triphosphate (ATP). Creatine can moderately regulate moisture in muscle tissues, expand muscle cross-section and improve muscle explosive power. Relevant experimental studies have verified that creatine has multiple beneficial effects on human body.

    Physiological Functions

    Creatine is synthesized from amino acids in the human liver and delivered to muscle cells through blood, and converted into creatine salt in muscle cells. It provides energy support for human muscle movement via ATP decomposition. It can automatically adjust moisture entering muscles, expand muscle cross-section and enhance muscle explosive power. A large number of in vivo and in vitro experiments have confirmed that creatine has the following functions:
    • Increase creatine content in skeletal muscle and improve energy reserve capacity
    • Regulate glucose metabolism and reduce lactic acid production
    • Improve intramuscular glycogen storage
    • Enhance antioxidant capacity of the body

    Applications

    Synthetic creatine is widely used as a nutritional supplement to assist skeletal muscle adaptation to high-intensity exercise and relieve physical fatigue. It can be used as a raw material for health-care preparations and compound health food ingredients with body conditioning effects.
    As a non-hormonal ingredient, creatine will not interfere with human endocrine system and cause dependence or adverse physiological reactions, so it is widely applied in health product industry. Natural creatine exists in a small amount in meat and fish products, which is insufficient for daily body conditioning needs, so supplementary intake of creatine monohydrate is commonly adopted to increase creatine content in skeletal muscle.
    Creatine can be converted into phosphocreatine under the action of creatine kinase, expanding the phosphocreatine energy metabolism pool. Supplementary intake of creatine monohydrate can optimize creatine metabolism, improve body growth status and tissue quality, and has great application potential in health nutrition and animal husbandry fields.
    In addition, creatine monohydrate can boost calcium uptake in sarcoplasmic reticulum, optimize the synthesis of myosin heavy chain and actin, support faster and stronger muscle contraction, help maintain lean body mass, and increase the cross-sectional diameter of muscle fibers, especially type II muscle fibers.

    Chemical Properties

    White crystalline or crystalline powder. Slightly soluble in water, insoluble in ethanol and ether. Hygroscopic under normal storage environment.

    Synthesis Method

    Raw material related CAS No.: 107-97-1, 7404-50-4, 6020-87-7
    1. Dissolve 196g (2.2mol) N-methylglycine (sarcosine) in 360g of water, heat the solution to 80℃ to form a homogeneous transparent solution.
    2. Rapidly add the above hot solution into the pre-prepared S-(carboxymethyl)isothiourea aqueous solution, control the feeding rate to keep the reaction temperature below 60℃.
    3. Stir and react the mixture continuously at 60℃ for 3 hours to ensure sufficient reaction.
    4. Stop heating after the reaction is completed, transfer the reaction system to an ice water bath and cool to 0-5℃ for crystallization.
    5. Filter to collect white crystalline products, wash the filter cake with a small amount of absolute ethanol to remove impurities, and dry to obtain 238g creatine monohydrate (white crystal, melting point: 294℃) with a yield of 80%.
    6. Extract the filtrate four times with 300ml chloroform, and combine all chloroform extract liquids.
    7. Recover chloroform by atmospheric distillation, and conduct vacuum distillation of the residual substance under 0.098MPa.
    8. Collect the distillate at 104-106℃ to obtain 156g mercaptoacetic acid with a yield of 85%.
