TRx‑0237 mesylate (LMTX, Hydromethylthionine) is a second‑generation tau‑protein aggregation inhibitor for laboratory research.

In‑vitro Research

TRx‑0237 (LMTX™) represents a purified form of methylene blue for research investigation. Methylene blue is a historically documented compound. In‑vitro assays indicate that 0.16 μM TRx‑0237 interferes with paired‑helical filaments (PHF) isolated from human brain tissue samples.

In‑vivo Research

In pre‑clinical animal model studies, TRx‑0237 showed dose‑dependent effects on learning performance in L1 mouse strain (over‑expressing truncated tau protein), and restored behavioral flexibility in water‑maze tests (minimum effective dosage: 9 mg MT/kg). For L66 mouse strain (over‑expressing full‑length tau‑P301S/G335D), the compound modulated motor‑learning performance (effective dosage: 4 mg MT/kg). It reduced the population of tau‑active neurons, prominently in hippocampus and entorhinal cortex of L1 animals, with more widespread observations in L66 strain.
