Eloralintide AMYR agonist 2883634-40-8

Eloralintide is an investigational, selective amylin receptor agonist primarily intended for weight management in adults who are obese or overweight (and have at least one weight-related comorbidity).
As a monotherapy, eloralintide has demonstrated significant efficacy in obese or overweight patients without type 2 diabetes. Results from a 48-week Phase II clinical trial (NCT06230523) showed weight reductions ranging from 9.5% to 20.1% among patients treated with various doses of eloralintide, significantly outperforming the placebo group (0.4%).
Eloralintide is also being evaluated as part of combination therapy—specifically in conjunction with tirzepatide, a dual agonist of the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors—for the treatment of obese or overweight patients with type 2 diabetes. This approach is currently being investigated in a Phase II clinical trial (NCT06603571).
As a selective amylin receptor (AMY1R) agonist, eloralintide functions by mimicking the activity of endogenous amylin; it promotes weight loss by delaying gastric emptying and enhancing satiety, thereby reducing caloric intake. This mechanism differs from that of mainstream GLP-1 receptor agonists (such as semaglutide) and represents a promising direction for next-generation weight-loss therapies. Eloralintide holds the potential to complement GLP-1-based drugs or offer a new treatment option for patients who are intolerant to GLP-1 medications.
